Analog (analogue)

Definition
An analog is a molecule built to closely resemble another, with small changes that improve how it works — often longer-lasting or more potent.
An analog is a molecular cover version. You start with something the body already uses — a hormone, a peptide — and make small, deliberate changes to it. The result is close enough to still fit the same receptor, but tuned to behave better as a medicine.
Think of it like reinforcing a paper cup. The original works fine but falls apart fast; swap a few materials and you get something that does the same job and survives far longer. That’s usually the goal with an analog: make it last.
Natural peptides often get destroyed in the body within minutes. Chemists get around that by tweaking the amino acid sequence, or bolting on a fatty acid chain, so the analog resists breakdown and sticks around. Semaglutide is an analog of the gut hormone GLP-1: nearly the same signal, but engineered from a few-minute half-life into a once-weekly drug.
Here is what that engineering looks like in semaglutide, a practical real-world example of an analog in modern drug design. It retains 94% sequence homology with human GLP-1, but a change at position 8 protects it from DPP-4, an enzyme that would otherwise cut the peptide apart quickly. A C18 fatty di-acid attached at position 26 also helps the molecule bind albumin, which slows clearance; together, these changes stretch its elimination half-life to about one week and support once-weekly dosing. The receptor signal remains GLP-1-like, while the pharmacokinetics, what the body does to the compound, are deliberately different, making semaglutide a clean example of an analog rather than a wholly new biological signal.
So when a compound is called “an analog of X,” read it as: it’s a purpose-built relative of X, made to be more stable, more potent, or more selective. Same neighborhood, better engineering. (British and scientific writing spells it “analogue” — same word.)
