Definition
Topical means applied to the skin surface — which is not the same as absorbed, since most peptides are too large to cross it passively.
The topical peptide meaning is simple: a peptide is applied to the skin in a cream, serum, gel, or patch rather than swallowed or injected. The phrase describes where the product goes, not how far the peptide travels. A topical peptide may stay on the surface, enter skin layers, or reach the bloodstream, depending on the exact molecule and formulation.
What does topical application mean?
Topical application means placing a substance on a particular spot on the body’s outer surface. The FDA distinguishes topical administration from transdermal delivery, which carries a substance through the skin toward systemic circulation. That distinction is small on a label and large in biology: “applied” reports an action; “absorbed” reports a result.
Topical vs injected is simpler. An injection puts a peptide beneath or through the skin barrier. A topical product must deal with that barrier first. Neither route proves that a peptide works for the claimed purpose; route and outcome are separate questions.
Do topical peptides absorb through skin?
Topical peptides can absorb, but the name alone gives no answer. Intact skin’s outer layer, the stratum corneum, strongly limits passive movement. The 500 dalton rule says molecules above roughly 500 Da rarely cross that layer passively. Many cosmetic peptides exceed that size, so “contains peptides” is not a penetration study in disguise.

How should topical peptide evidence be graded?
Topical peptide evidence should be graded in three separate steps: first, whether the exact peptide can cross intact skin; second, whether the tested formulation delivers enough to the intended skin layer; and third, whether a controlled human study found the claimed result. Cell activity clears none of those delivery hurdles by itself.
GHK-Cu is a useful concrete example because it appears in both skincare and injectable discussions. “Topical GHK-Cu” identifies the route, not an absorption percentage, and evidence for a serum cannot be borrowed from an injection.
