Pulsatile release

Definition

Pulsatile release means a hormone comes out in bursts separated by quieter periods, rather than flowing at a constant rate.

Pulsatile release means a hormone comes out in bursts rather than a constant trickle. For growth hormone (GH), levels rise sharply, fall back, and rise again, with the most reproducible adult pulse often arriving soon after sleep begins. That timing is physiology; claims that a peptide preserves it need a separate evidence check.

What does pulsatile mean?

The plain-English pulsatile meaning is “on, then mostly off, then on again.” A pulse has a peak and a quieter interval after it. Steady release keeps exposure closer to a continuous baseline. The same total hormone output can therefore have a very different shape across the day: mountain peaks versus a raised waterline.

When do growth hormone pulses happen?

Growth hormone pulses occur across the day and night, but the most reproducible adult burst follows sleep onset and the first period of slow-wave sleep. A human physiology review describes that sleep-onset surge as often the adult’s major GH output (Van Cauter and Plat, 1996). Age, sex, sleep, exercise, and body composition can all change GH pulsatility, so no single graph represents everyone.

Do sermorelin and ipamorelin preserve GH pulsatility?

Sermorelin and ipamorelin prompt the pituitary to release GH instead of supplying GH directly, which leaves the gland and feedback loop involved. Ipamorelin has direct human pharmacology behind the “pulse” wording: one study measured a single episode of GH release that peaked and then declined toward negligible levels (Gobburu et al., 1999).

Sermorelin’s pulse-preservation claim is less direct. Calling its GHRH-receptor mechanism “more physiologic” is reasonable mechanistic reasoning, but it is not proof from a head-to-head trial that preserving pulses improves sleep, recovery, body composition, or safety. What does pulsatile vs steady mean for CJC-1295 DAC?

CJC-1295 with DAC creates prolonged exposure rather than one brief GH event. In healthy adults, the DAC form raised mean GH for six days or more and IGF-1 for 9–11 days after one injection (Teichman et al., 2006). Calling that a steadier GH “bleed” is useful shorthand, not a trial finding that levels become perfectly flat.

The practical reading across the growth-hormone peptide family is simple: short GH peaks are measured facts for natural secretion and ipamorelin; better outcomes from “preserving” those peaks remain a hypothesis. Hormone pattern and clinical benefit are related questions, not interchangeable answers.
