MGF peptide is a 24-amino-acid fragment linked to the exercise-responsive IGF-1Ec transcript, but an injected fragment has never been shown to build or repair muscle in people. Cell studies conflict, PEG-MGF has no human exposure data, and neither form is FDA-approved. The honest evidence tier is in-vitro and mixed.
What is MGF peptide?
MGF peptide is a synthetic copy of the 24-amino-acid E-domain at the tail of IGF-1Ec, an alternatively spliced version of the IGF-1 gene. The verified research sequence is YQPPSTNKNTKSQRRKGSTFEEHK. “Mechano growth factor” can mean the full IGF-1Ec precursor, its messenger RNA, or this isolated E-peptide. Those are related, but they are not interchangeable.
That naming problem drives much of the hype. Muscle can increase IGF-1Ec gene expression after mechanical loading without proving that the free E-peptide circulates as a hormone or that a synthetic vial reproduces the same biology. MGF sits in the broader growth-hormone and IGF-1 family, but the isolated fragment does not have a confirmed receptor.
Does MGF peptide work for muscle growth or repair?
MGF peptide has conflicting cell-culture results and no human treatment evidence for muscle growth, strength, or recovery. A 2011 study reported that the 24-aa E-peptide extended the proliferative life span of muscle progenitor cells from neonatal and young-adult donors, delayed cellular aging, and changed fusion behavior. Cells from older adults did not show the same proliferation result (Kandalla et al.).
Then came the test vendor pages tend to misplace. Fornaro and colleagues tried to reproduce the satellite-cell story across C2C12 mouse myoblasts, primary human skeletal-muscle myoblasts, and primary mouse muscle stem cells. At concentrations up to 500 ng/mL, MGF did not increase proliferation, delay differentiation, or activate the signaling response they tested. Mature IGF-1 and full-length IGF-1Eb did produce responses in the same systems (Fornaro et al.).
That leaves MGF peptide results at mixed in-vitro evidence, not human proof. The disagreement is the result, not an inconvenient footnote.
Does exercise really increase mechano growth factor?
Mechano growth factor gene expression does rise after resistance exercise, but that is a different claim from injected MGF working. In a human muscle-biopsy study, IGF-1Ec messenger RNA was 1.7 times baseline at 24 hours and 1.5 times baseline at 48 hours after active recovery; after cold-water immersion, it reached 2.1 times baseline at 24 hours (Fuchs et al.).
Messenger RNA is the cell’s instruction sheet. Detecting more copies after training shows that muscle changed its IGF-1 splicing program; it does not show how much free 24-aa peptide was made, whether the peptide entered blood, or whether injecting a synthetic version adds muscle. The natural signal is real. The marketed inference is several steps ahead of it.
What is PEG-MGF, and is it better studied?
PEG-MGF is MGF attached to polyethylene glycol, a chemical modification intended to slow breakdown, but it has even less direct evidence than the unmodified fragment. “Longer lasting” is a design goal, not a clinical result. Published human pharmacokinetic, dose-ranging, efficacy, and safety trials for peg-mgf are absent.
FDA states that it found no human exposure data for drug products containing PEG-MGF by any route. FDA also flags possible immune reactions, peptide-related impurities, and difficulty confirming the active ingredient (FDA compounding safety review). PEGylation may change how a molecule behaves; it cannot be assumed to rescue a disputed mechanism.
What MGF peptide dosage has research established?
No MGF peptide dosage has been established for people. The 500 ng/mL figure from Fornaro is a concentration applied to cells in a dish, not an injection dose. Online microgram schedules are community or vendor protocols, not doses validated by a human trial, so this page does not convert them into a recommendation by repetition.
The same limit applies to timing, injection location, cycling, and stacking. Without human pharmacokinetics, even the basic questions—how long the unmodified peptide lasts, how PEG-MGF distributes, and what exposure reaches muscle—remain unanswered. Is MGF peptide safe? Side effects and risks
MGF peptide safety in humans is unknown because there are no controlled human exposure studies to count adverse events. That means “few reported side effects” would be empty reassurance: no denominator, no safety rate. FDA’s PEG-MGF review specifically raises immunogenicity and impurity-characterization concerns, while acknowledging that it lacks enough information to know whether the drug would harm people.
Research-market injections add separate risks from incorrect identity, concentration, sterility, and contaminants. Growth signaling also makes off-target tissue effects a reasonable research question, but no human MGF dataset can quantify that risk. The correct side-effect list is not a row of guesses; it is an evidence gap.
Is MGF peptide FDA-approved or banned in sport?
MGF peptide is not FDA-approved, and WADA prohibits mechano growth factors at all times. FDA’s May 14, 2026 compounding document places unpegylated mechano growth factor in Category 3, meaning it was nominated for 503A compounding without adequate support. PEG-MGF remains under review for a future advisory-committee discussion, which does not make it approved or endorsed.

MGF vs IGF-1: what is the practical difference?
MGF vs IGF-1 is a fragment-versus-growth-factor comparison, not two interchangeable names. Mature IGF-1 is a 70-amino-acid hormone with a known receptor and an FDA-approved recombinant form, mecasermin, for a narrow pediatric indication. MGF is the isolated 24-aa E-domain with disputed cell activity and no approved use.
IGF-1 LR3 is different again: an engineered 83-amino-acid IGF-1 analogue that activates the IGF-1 receptor but lacks human muscle-building trials. IGF-1 DES is another truncated IGF-1 analogue, not another name for MGF.
