P021 is a synthetic neurotrophic peptide — a small, chemically modified fragment built to boost brain growth-factor signaling — that has improved memory in mouse models of Alzheimer’s disease. P021 is not FDA-approved and has never been tested in a human trial. The rodent science is promising; the human proof does not exist yet.

What is P021?

P021 is a lab-made peptidomimetic — a short peptide-like molecule engineered from a four-amino-acid core (aspartate–glycine–glycine–leucine, or “DGGL”). Chemists acetylated one end and capped the other with an adamantane group (a rigid, cage-shaped carbon structure) so the molecule survives digestion and can cross into the brain, which most peptides can’t. Its formula is C27H42N6O8, its molecular weight is about 578.7, and its CAS number is 1246751-68-7 (PubChem CID 56599151). P021 was created as a neurotrophic (“nerve-growth-supporting”) research compound, and it is sometimes written P21 or P-21. It is not a medicine you can be prescribed and not a supplement you can legally buy — in the United States P021 exists only as a research chemical.

How does P021 work?

P021’s proposed mechanism, drawn entirely from animal work, centers on brain-derived neurotrophic factor (BDNF) — a signaling protein that acts like fertilizer for neurons, helping them survive, connect, and form new memories. In rodents, P021 appears to raise BDNF activity and encourage neurogenesis, the birth of new neurons in the hippocampus (the brain’s memory hub). What the community reports

Outside the lab, P021 is far more obscure than headline peptides like BPC-157, and online discussion is thin. These accounts are anecdotal, and they sit next to a none-in-humans reality: they are not controlled data, they suffer from survivorship bias (people who feel nothing rarely post), and no one is measuring whether anything is actually happening in the brain. Is P021 safe? Side effects
